Why Can You Choose Salivary DNA For Whole Genome Sequencing (WGS)? Time For A New Perspective!

“Whole genome sequencing (WGS) is a laboratory procedure that determines the order of bases in the genome of an organism in one process” – CDC

WGS is being increasingly used for both research and clinical applications to study the role of genetic variation in understanding human diseases. As the cost of sequencing continues to decrease, WGS is emerging as a new player in the molecular disease diagnostics niche.

The standard sample of blood-derived DNA is currently used for WGS.

Well, clinical situations can vary from daily common chores to critical and rare scenarios. Isn’t it?

Think of a patient who has a significantly reduced number of white cells in the blood (leucopenia) for any disease reason or who has been recently exposed to blood transfusions. Another clinical situation could be a patient presenting with hematopoietic stem cell transplantation from an allogeneic* donor.

In none of the above situations, blood can be used as a DNA extraction sample. What could be the best alternative?

Yes! It is ‘SALIVA”!

The use of salivary DNA for WGS is not so popular or not well known now. Though it comes with a few limitations such as lower DNA yield, microbial contamination if not properly collected, etc., salivary DNA can be one of the safest choices for WGS.
